shut off water main

Should you shut off water main and water heater when on vacation . Also if there is water line under slab before it reaches the water main, how can you ensure that there is no water leak from there. Will a water leak detection come in handy?

HOw long will the vacation be, and do you have anyone checking the house periodically. It is safer to turn the water supply and water heater off for extended periods, but it can have adverse effects in some situations. You cannot "ensure" that there is no leak ANYWHERE, unless the piping is checked. If you know there is a leak, then a leak detection company is recommended tp find out where it is.
