Cost to install tub drain assembly

We have an antique claw footed tub with a leaky drain. A plumber came to give us an estimate and said that to replace the entire drain assembly (drain + overflow) would be $1,200.

Home Depot and other places list the assembly at around $150, so the cost of installation is about $1,050. The going rate for plumbers is around $175 here, so this works out to be about 6 man hours of work. Is this reasonable?

probably not, but we do not know the situation. If it is all exposed then it would normally only take an hour or at most two to do it. but you are probably dealing with a "flat rate" plumber who uses a price book to determine your cost, regardless of how long it would actually take.
