You can do anything you want to as far as a drain size is concerned. The only time it becomes a factor is when you install a 1 1/2" drain, cover it up with a finished wall, and then find out that it overflows everytime the washing machine drains. Why would you run that risk, just to save a few pennies on materials? For your purposes, you can drill a 2 9/16" hole in the 2x4, and even notch the front of it so you can insert a full piece without couplings, and it will not reduce the structural stability of your project by one iota.
