If drain has a rubber grommet do I need plumbers putty?

Pulled into it to clean out and it had putty originally installed.

Usually plumbers putty goes on the sink side and the rubber washer goes underneath. If the sink is acrylic might want to check if the plumbers putty is compatible.

Also use putty on the threads below to stop water from following the threads down.

Pay attention to make sure that you get the right drain-- with an overflow path, unless you use a vessel sink with no overflow. If you use a vessel sink with no overflow, understand that may not function as well as a sink with overflow.
