Attached is the faucet Knob without the plastic handle. 1) How does the Knob come off? 2) Do I need to turn the water off to replace the handles? The old ones cracked and fell off. I bought a universal handle kit. How do I replace them? Do you think these will work?

That plastic knob is a cheap, poor quality, aftermarket universal thing. The spline assembly should come off the stem if it has a screw in the center. otherwise, maybe the spline is crushed on. All junk.
