Can one hole have the supply, drain and power (not hard wired)through it or does it need to be separate holes.

I think you are asking if you could use a big hole inside of the sink cabinet instead of 3, like the one drawn in red.

You can use as many, or few, holes as you want to. A single hole will usually be near the bottom, not up high like that "red oval".
