Check local codes BUT normally YES you can have one 4" roof penetration.

You should also count the fixture units connected to the vent line your going to run on the horizontal and measure the developed length to make sure your with in the proper size of venting.
Do not forget to pitch the vent up from the fixtures being served so if any condensate should occur it will flow back to the drain it is servicing
