With the exception of traps, there is no water in a properly installed drain pipe when it is not actually draining water. There is absolutely no connection between your supply and the drain. To check for a hidden leak, with all water outlets off, check the water meter. If the dial moves, then you have a leak. If the dial remains stationery, there's no leak. A vent connects to the drain that it is venting and goes to the outside through the attic. I have no idea what that open pipe is for, but it is obviously connected to the drain, and it does not have a trap or the trap is dry. In either case, it should not be open. Get a cap for it then have your plumber check your system as soon as possible.
