Is it necessary to use a hydraulic separate or low-loss header in a system with a low head loss boiler; a single variable speed-constant pressure circulator; and a home-run distribution system with manifolds? Boiler I'm looking to use is the Westinghouse WBRU fire tube modcon, which has low pressure losses at the boiler at my design flow rates.

It's not usually necessary with fire tube boilers (or even most others, if you set up the flow control properly), which is why HTP's marketing harps on the "No Primary/Secondary Piping Needed") for their UFT series, which are identical (under the paint) to the Westinghouse fire tube boilers.
